The school has a long history and ample space. The school was founded on November 21, 1934 and is located in Nanning, the capital of Guangxi Zhuang Autonomous Region. Since its establishment, the school has moved seven times and changed its name seven times. The predecessor of the school was Guangxi Provincial Medical College. Due to the turbulent situation, the school moved to Guilin in 1940 and moved back to Nanning from Guilin in July 1954. After the founding of New China, the school was renamed Guangxi Medical College in November 1949; in 1952, it was directly led by the Central and South Ministry of Health entrusted by the Central Ministry of Health; in April 1953, the Central Ministry of Health approved the name change to Guangxi Medical College; in May 1996, the Ministry of Education approved the name change to Guangxi Medical University.

The school is one of the 22 earliest medical schools in the country, one of the eight medical schools in the country that first recruited foreign students, students from Hong Kong, Macao and Taiwan, and overseas Chinese students. It is one of the first 30 universities approved by the Ministry of Education to recruit undergraduate clinical medicine majors (taught in English) and is the center of medical education, medical research, clinical medicine and preventive health care in Guangxi. In May 2020, it became the only medical school in Guangxi jointly built by the Autonomous Region People's Government and the National Health Commission. In 2021, it became a university that the Autonomous Region's "14th Five-Year Plan" focuses on supporting the construction of a domestic "double first-class" university.

The school currently has four campuses: Nanning-Main Campus, Nanning-Wuming Campus, Yulin Campus, and International Medical Education Exchange and Research Center (Wuxiang Campus, under construction).
